These sliders are called 'faders'.
They affect the output signal of the track.
As you see, fader for each track is visible in the mixer strip (at the left side), so you may place the processing before the loudness is adjusted - before the fader - and after the loudness is adjusted - after fader. On the picture your AUDynamicsPro plugin is placed into post-fader position, so it processes already adjusted by the volume signal. I think if you want to adjust the loudness of processed signal, you should better place all plugins into pre-fader position. Post-fader position can be used for such processors like limiters and sends (for example, send on a reverb bus, the signal for send to reverb bus is better to be affected by the fader).
