I tried Openbox but the Mixbus GUI wouldn't even appear! I realise I was using the default config so maybe there is something I would need to tweak. Have you tried another WM? JWM is a very lightweight, easy test (I use it for some games that don't like Compiz).

JWM I have and I've got almost all the desktops installed now and none of them will load it. Maybe it's something I did wrong because mine did not come with an installer so I just copied it into the VST directory. I just copied the VST into ~/.vst and the "standalone" elsewhere. Both work the same. Have you tried the standalone? I wouldn't call that a bad habit I also do the same. Try running ldd on the .so file (the plugin file) in a terminal.
It will print all the dependencies that the plugin has to libraries on your computer. In displayed list, check for any entry that failed to resolve. I don't remember the exact wording but it should be apparent that something is missing.
If there is a dependency missing, the plugin will likely not work.
I had this problem with the Aural Verb in the beginning. Basically it made a few assumptions that support libraries already were installed on your system. It can usually be fixed by installing the missing library, which I could in that case. I bug reported it to them also to hopefully require fewer dependencies. It's kept working here and I haven't checked if they improved it.

The aural verb causes the Mixbus 32C to mute randomly. I had it on Mixbus and suddenly the whole session went silent. When Aural verb put bypass the session started playing again. So this was just a drum reverb in one Mixbus. AVLinux MXE latest version.
