Full message after clicking on that tab under the Assistant (for 64bit apps) was:

regsvr32: Failed to load DLL 'wineasio.dll'
Do I not need to run this?

You need to run this if you want to run a Windows Audio application with ASIO drivers, is that what you're doing?
Also what version of Wine-Staging are you using? If you've updated to the latest version 6.7 my guess is the Wine guys have broken WineASIO. I am using Wine-Staging 6.2 here and WineASIO is working as expected. Wine has been very stable for a long time but in the past 2 months it has broken support for Windows VST Audio Plugins at least 3 times.. You can downgrade Wine if you need to and I can help you with that.
